In an attempt to select potential biocontrol agents against Pythium spp. and Rhizoctonia spp. root pathogens for use in soilless systems, 12 promising bacteria were selected for further investigations. Sequence analysis of the 16S rRNA gene revealed that three strains belonged to the genus Enterobacter, whereas nine strains belonged to the genus Pseudomonas. In in vitro assays, one strain of Pseudomonas sp., Pf4, closely related to Pseudomonas protegens (formerly Pseudomonas fluorescens), showed noteworthy antagonistic activity against two strains of Pythium aphanidermatum and two strains of Rhizoctonia solani AG 1-IB, with average inhibition of mycelial growth >80%. Strain Pf4 was used for in vivo treatments on lamb’s lettuce against R. solani root rot in small-scale hydroponics. Pf4-treated and untreated plants were daily monitored for symptom development and after two weeks of infection, a significant protective effect of Pf4 against root rot was recorded. The survival and population density of Pf4 on roots were also checked, demonstrating a density above the threshold value of 105?CFU?g?1 of root required for disease suppression. Known loci for the synthesis of antifungal metabolites, detected using PCR, and draft-genome sequencing of Pf4 demonstrated that Pseudomonas sp. Pf4 has the potential to produce an arsenal of secondary metabolites (plt, phl, ofa and fit-rzx gene clusters) very similar to that of the well-known biocontrol P. protegens strain Pf-5.  相似文献

Rhizoctonia solani is one of the most problematic soil-borne pathogenic fungi for several crop cultures worldwide. This study highlights the effectiveness of high-antagonistic Streptomyces rochei strain PTL2, isolated from root tissues of Panicum turgidum, in controlling the R. solani damping-off and growth promotion of tomato (cv. Marmande) seedlings. The isolate PTL2 was characterised for in vitro biocontrol and plant growth-promoting traits. It exhibited remarkable positive results in all trials, including production of hydrogen cyanide, siderophores, 1-aminocyclopropane-1-carboxylate deaminase and phytohormones, chitinolytic activity and inorganic phosphate solubilisation. PTL2 spores were formulated as wettable talcum powder, sodium alginate pellets and sodium alginate-clay pellets. Their abilities in the biocontrol of R. solani and plant growth promotion were investigated in autoclaved and non-autoclaved soils. Talcum powder and sodium alginate pellets significantly reduced the damping-off severity index compared to a positive control. The talcum powder exhibited the highest protective activity, reducing the disease incidence from 89.3% to 14.1%, whereas chemical seed treatment with Thiram® provided a disease incidence of 16.7%. Furthermore, the talc-based powder formulation resulted in greatest increases in the root length, shoot length and dry weight of seedlings. The interesting biocontrol potential and growth enhancement of tomato seedlings open up promising perspectives for the possible application of talcum powder formulation based on PTL2 spores in crop improvement.  相似文献

Baseline sensitivity and efficacy of thifluzamide in Rhizoctonia solani   总被引:1,自引:0,他引:1  
Thifluzamide is a SDHI (succinate dehydrogenase inhibitor) fungicide, which interferes with succinate ubiquinone reductase in the mitochondrial electron transport chain of fungi. Presently, jinggangmycin is the major fungicide extensively used for the control of rice sheath blight caused by Rhizoctonia solani and resistance to jinggangmycin was first reported to occur in China. A total of 128 isolates of R. solani from Anhui Province of China were characterised for the baseline sensitivity to thifluzamide. The isolates were very sensitive to thifluzamide and the baseline sensitivity curve was unimodal with an average EC50 value of 0.058 ± 0.012 µg mL?1. However, EC50 values of boscalid (another SDHI fungicide) for inhibition of mycelial growth of 22 arbitrarily selected R. solani isolates ranged from 1.89 to 2.68 µg mL?1. Thifluzamide applied at 110 µg mL?1 exhibited excellent protective and curative activity against rice sheath blight and provided 81.1–91.0% protective or curative control efficacy. In field trials in 2010 and 2011, control efficacies of thifluzamide at 82 g.a.i ha?1 15 and 30 days after second application were 84.2% and 86.7%, respectively, suggesting excellent activity against sheath blight. There was a statistically significant difference in the efficacy between thifluzamide and boscalid or jinggangmycin. These results suggested that thifluzamide should be a good alternative fungicide to jinggangmycin for the control of rice sheath blight.  相似文献

枯草芽胞杆菌EBS05是从樟树中分离的1株对多种植物病原真菌具有拮抗作用的内生细菌。以小麦纹枯病菌为靶标菌,通过单因素试验和正交设计试验对其发酵条件进行了优化。结果表明,内生细菌EBS05适宜的发酵培养基主要营养成分的组成和配比分别为可溶性淀粉3%、蛋白胨2%、NaCl 0.25%。最佳发酵条件为:初始pH5~9,最适温度34℃,装液量25 mL/250 mL三角瓶,接种量3%,发酵时间72 h。  相似文献

The major problem in agriculture is that of the fungal pathogens. In this era, where biological control is at focus, and is the centre of crop protection as well as environmental protection, the synthesis of new bio bodies is of utmost need. Fungicides available in the market, though of potential, are pathogen specific and highly pollutive. An attempt was made to raise polyclonal antibodies against Aspergillus niger. Following the particular standardised immunisation schedule, regular injections and periodic tapping were carried out. The IgG purified was used to check cross-reactivity with different crop fungi like Fusarium solani, Fusarium oxysporum f. sp. cicer (FOC), Rhizoctonia bataticola, Botryodiplodia theobromae, Curvularia sp., Alternaria porri and also with Aspergillus niger by two different methods. Liquid test media and the radial growth inhibition test performed in solid media were used to check the inhibition of fungi and cross-reactivity. The results were subjected to analysis of variance (ANOVA) by using Tukey's test at the significance level of p < 0.05. The antibodies were active against all the fungi for more than 15 days except for A. niger in which from the seventh day onwards spore germination was observed. The probable role of antibodies to detect the common antigenic molecule that may be present in all the tested fungi and their role in inhibiting these fungi is discussed.  相似文献

Rhizoctonia bataticola produced oxalic acid in vitro and in vivo during pathogenesis of patato tuber. Polygalacturonase (PG) was also detected in culture filtrates of the rot-causing organism. Interaction of tomato roots with Trichoderma virens TRS106 provided protection against Rhizoctonia solani-induced disease. In tomato, plants inoculated with R. solani disease symptoms were observed on the roots as brown, necrotic lesions. These symptoms were limited on plants treated with TRS106 and inoculated with R. solani. It was shown that TRS106 did not directly inhibit Rhizoctonia growth in in vitro test. The tested Trichoderma isolate stimulated systemic defence responses in tomato plants, by activating defence enzymes including guaiacol peroxidase (GPX), syringaldazine peroxidase (SPX) and phenylalanine ammonia lyase (PAL). Simultaneously, it enhanced accumulation of phenolics and hydrogen peroxide (H2O2) accompanied by decrease in lipid peroxidation in the leaves. HPLC analysis indicated remarkable increases in the concentrations of 22 phenolics in the leaves of Trichoderma-treated tomato, both uninoculated and inoculated with R. solani. Some of the phenolics were present in a free form, the others were accumulated in a bound form as glycosylated conjugates belonging to phenylpropanoids, hydroxybenzoic and cinnamic acid derivatives and flavonoids. Several of the detected phenolics: ferulic and salicylic acids, pyrocatechol and hesperetin were strongly toxic to R. solani in plate tests. The systemic mobilisation of phenolic metabolism might be an element of tomato defence response positively involved in biocontrol of R. solani by TRS106. Based on the results, T. virens TRS106 may have potential to develop a new biofungicide for integrated management of R. solani-induced disease.  相似文献

During two growing seasons (2008 and 2009), the associations of Rhizoctonia root rot (RRR) with a number of soil properties were determined at different growth stages in 122 commercial bean fields in Zanjan, Iran. Mean RRR incidence at a level of 4–25% sand content was lower than that at 45–65% level. Damage by fly puparia had no significant effect on RRR incidence and occurrence. A greater RRR incidence was detected in field soils treated with fungicides compared with non‐treated soils. A lower RRR incidence was associated with the highest level of soil organic matter (1.2–1.8) compared with the lowest level, 0.4–0.8. The highest RRR incidence corresponded with no rhizobial nodulation compared with highly nodulated bean roots. RRR incidence was negatively correlated with soil silt and organic matter content at R6–7 and R9 growth stages. RRR‐affected fields were recognized with a greater soil pH (V3) and sand content (R9), and a lower silt (R9) and organic matter content (R6–7 and R9) in comparison with RRR‐free fields. Loadings and linear regressions between RRR incidence and principal component scores indicated that the most effective soil characteristic linked to the disease was silt at V3, sand at R6–7 and organic matter at R9 stage. This new epidemiological information extends our knowledge of the bean–RRR–soil interaction on a regional basis.  相似文献

Abstract

We developed a modified staining technique using acridine orange to stain the nuclei of Rhizoctonia solani. Acridine orange solution was prepared in acetic acid buffer, pH 7.2. Staining for 15 min was critical for observing the nuclei. All of the isolates were found to be multinucleated. The nuclei appeared bright green with light orange background.
